How it works.

Semaglutide is a 31-residue GLP-1 analog modified at position 8 (Aib) to resist DPP-4 degradation and conjugated at K26 to a C18 fatty diacid linker that promotes albumin binding. Half-life is approximately 165 hours — roughly 100× that of native GLP-1. In binding studies it engages the GLP-1 receptor with sub-nanomolar affinity, producing downstream effects on insulin secretion, glucagon suppression, gastric emptying, and central satiety pathways.
